COLORECTAL DISTENSION-INDUCED EMG IN THE ABDOMINAL MUSCLES AS ONE OF THE PARAMETERS FOR THE VISCERAL PAIN

Abstract
Colorectal distension (CRD) have been used as a stimulus for the visceral pain study for it can be done easily, repeatedly
and almost harmlessly. But simple observation of behavioral sign such as writhing movements seems like not sufficient or convenient for estimating pain. Nowadays several research groups are using electromyography (EMG) of corresponding skeletal muscles to the visceral tissues for estimation of visceral pain intensity, based on viscero-skeletal reflex mechanisms. We tried this strategy for the colorectal inflammatory pain assessment. A balloon for CRD was inserted through the anus into male Sprague-Dawley rats, in which two wires were installed as electrodes into the abdominal muscle at least 3 days before experiments. The ends of the wires, exposed at the occipital region, were connected to EMG recording system and EMGs were recorded. CRD pressure was fixed to 80 mmHg and a series (5 times, every 2 minutes) of CRD was delivered 3 times every 30 minutes before and after zymosan or capsaicin treatment. After zymoan (2.5%) treatment, the EMG activities of the abdominal muscle to CRD were enhanced significantly stimuli, comparing to EMG before the treatment, and the enhanced responsiveness is continued until 4 hrs after. Capsaicin (0.2%) treatment decreased the EMG activities to CRD stimuli, but the EMG activities during interva periods between DRD were increased, intermittently. These results suggest that EMG in the abdominal muscles following CRD should reflect the intensity of visceral pain and could be used as one of the parameters for visceral pain intensity.
